Cagrilintide Reconstitution Math
Reconstitution math starts with the amount contained in the vial and the liquid volume used in the calculation. Dividing the vial amount by the liquid volume gives the resulting concentration.

What is Cagrilintide?
Cagrilintide is a synthetic long-acting analog of amylin, a pancreatic hormone involved in satiety and meal-related signaling. It is being developed by Novo Nordisk as an investigational obesity medicine. Cagrilintide is not currently an FDA-approved drug.
How does Cagrilintide work?
Cagrilintide is designed to activate amylin-receptor signaling, which can influence satiety, food intake, and gastric emptying. Its mechanism is different from GLP-1 receptor agonists such as semaglutide, which is why the two mechanisms have also been studied together in CagriSema.
What has human research shown?
A multicenter randomized Phase 2 trial in 706 adults with overweight or obesity found greater mean weight reduction with once-weekly cagrilintide across the studied dose groups than with placebo over 26 weeks. Gastrointestinal adverse effects and injection-site reactions were among the most common adverse events. Phase 2 findings established direct human efficacy evidence but did not create an approved treatment.
Cagrilintide is now in Phase 3 development
Novo Nordisk has advanced cagrilintide into the RENEW Phase 3 program for weight management. ClinicalTrials.gov records show active Phase 3 studies, including RENEW 2 in adults with overweight or obesity and type 2 diabetes. This means cagrilintide has moved beyond early clinical research but remains investigational while those programs continue.
Cagrilintide vs CagriSema
Cagrilintide and CagriSema are not the same intervention. Cagrilintide is the amylin analog alone. CagriSema is a fixed-dose combination of cagrilintide and semaglutide. Results from CagriSema trials cannot automatically be attributed to cagrilintide alone because both active components contribute to the combination.
Safety context
In cagrilintide trials, gastrointestinal adverse effects such as nausea, constipation, and diarrhea have been common, along with injection-site reactions. Because the drug remains investigational, its long-term benefit-risk profile is still being defined through larger and longer clinical programs.
