Synthetic Pineal Tetrapeptide
Epitalon, also called Epithalon or Epithalone, is a synthetic tetrapeptide made from alanine, glutamic acid, aspartic acid, and glycine. It is widely discussed in longevity, telomere, circadian-rhythm, and sleep communities, but most evidence remains preclinical, cell-based, or limited human research. This page explains the current evidence, safety and regulatory status, common claims, and educational reconstitution math.

Epitalon Reconstitution Math
Reconstitution math starts with the amount contained in the vial and the liquid volume used in the calculation. Dividing the vial amount by the liquid volume gives the resulting concentration.
Example vial amount
10 mg
| Liquid Volume | Calculation | Concentration |
|---|---|---|
| 1 mL | 10 mg ÷ 1 mL | 10 mg/mL |
| 2 mL | 10 mg ÷ 2 mL | 5 mg/mL |
| 3 mL | 10 mg ÷ 3 mL | 3.3333 mg/mL |
This table demonstrates concentration math only. The liquid volumes shown are examples for comparison and are not preparation recommendations.

What is Epitalon?
Epitalon is a four-amino-acid peptide with the sequence Ala-Glu-Asp-Gly, often abbreviated AEDG. It was developed from research on peptide preparations associated with the pineal gland. Epitalon is not an FDA-approved medication in the United States, and products marketed online should not be assumed to have established pharmaceutical quality, safety, or effectiveness.
Epitalon vs Epithalamin
Epitalon and Epithalamin are related in the historical research literature but are not the same substance. Epithalamin is a complex peptide preparation derived from pineal tissue, while Epitalon is the specific synthetic tetrapeptide Ala-Glu-Asp-Gly. Some older longevity claims are based on Epithalamin studies and should not automatically be attributed to Epitalon itself.
Why is Epitalon discussed for longevity?
Epitalon became popular in longevity discussions because laboratory studies reported effects involving telomerase activity, telomere length, chromatin structure, circadian signaling, and lifespan in some animal models. These findings are scientifically interesting, but they do not establish that Epitalon extends human lifespan or prevents age-related disease.
Epitalon and telomerase
Early cell-culture studies reported that Epitalon increased expression and activity of telomerase in human fibroblasts and was associated with telomere elongation. In one study, treated aging fibroblasts continued dividing beyond the point at which control cells stopped. This was an in-vitro finding and should not be interpreted as evidence that administering Epitalon lengthens telomeres or extends lifespan in people.
What does newer telomere research show?
A 2025 Biogerontology study again found that Epitalon increased telomere length in multiple human cell lines and reported changes involving hTERT expression, telomerase activity, and alternative lengthening of telomeres pathways. The study strengthens the evidence that Epitalon can influence telomere biology in cultured cells, but it still does not establish a clinical anti-aging benefit in humans.
Does Epitalon extend human lifespan?
There is no modern, well-controlled clinical trial demonstrating that Epitalon extends human lifespan. Some frequently cited long-term gerontology studies involved Epithalamin rather than the synthetic Epitalon tetrapeptide. Animal and cellular longevity findings are hypothesis-generating and cannot establish a human lifespan effect.
Epitalon, melatonin, and circadian rhythm
Pineal-peptide research has reported changes in melatonin-related signaling. A randomized placebo-controlled study evaluated sublingual AEDG peptide in women ages 40 to 50 who worked mainly night shifts and had low melatonin-metabolite levels. After 20 days, urinary 6-sulfatoxymelatonin increased in the peptide group. The study assessed a melatonin metabolite and clock-gene expression rather than demonstrating treatment of insomnia.
Does Epitalon improve sleep?
Epitalon is commonly marketed for sleep because of its association with pineal function and melatonin. However, FDA's 2026 review concluded that available evidence does not support the effectiveness of Epitalon free base or Epitalon acetate for insomnia. The available studies do not establish that Epitalon improves sleep onset, sleep duration, sleep architecture, or clinically diagnosed insomnia.
What have animal studies shown about circadian signaling?
Studies in older rhesus monkeys reported increased evening melatonin and altered cortisol rhythms after Epitalon exposure. FDA noted important limitations: sleep behavior and electroencephalographic activity were not measured, dose-response relationships were not established, and the functional relevance of the hormone changes remains uncertain.
What is known about Epitalon safety?
Human safety is not well characterized. In its 2026 compounding review, FDA stated that it had not identified clinical studies that adequately assessed the safety of Epitalon-related bulk drug substances in humans. FDA also cited potential immunogenicity concerns related to aggregation, peptide impurities, and uncertainty around active-ingredient characterization, particularly for proposed injectable use.
Epitalon and cancer-related concerns
Because some laboratory studies report telomerase activation, questions have been raised about whether Epitalon could influence cancer biology. Telomerase is active in many cancers, but there is no evidence establishing that Epitalon causes cancer in humans. FDA's 2026 review noted that available data are insufficient to perform a full assessment of human carcinogenic potential, so this area remains unresolved rather than proven safe or harmful.
FDA and compounding status
Epitalon is not a component of an FDA-approved drug and does not have an applicable USP or National Formulary drug-substance monograph. In July 2026, FDA evaluated Epitalon free base and Epitalon acetate for possible inclusion on the Section 503A Bulks List and proposed that neither be included. FDA cited limited characterization, insufficient human safety data, lack of evidence supporting effectiveness for insomnia, and product-quality concerns.
Why product identity matters with Epitalon
FDA has noted inconsistent naming of materials sold under the common name Epitalon, including salts and related forms that may not represent the same active moiety. This matters because a label saying 'Epitalon' does not by itself establish chemical identity, purity, concentration, sterility, or equivalence to material used in published research.
Research Epitalon vs an approved medication
Online research products are not equivalent to FDA-approved pharmaceuticals. Epitalon has no FDA-approved product with standardized labeling, manufacturing controls, validated human dosing, established indications, or an approved safety profile. Claims involving anti-aging, longevity, sleep, or disease prevention should therefore be separated from established medical evidence.
Epitalon reconstitution math
Reconstitution calculations depend only on the labeled amount in a vial and the liquid volume entered. As a mathematical example, a 10mg vial combined with 2mL of liquid produces a concentration of 5mg per mL. BACToBasics uses examples like this to demonstrate concentration, liquid-volume, and U-100 syringe-unit conversions rather than to recommend an Epitalon preparation or dosing protocol.

Epitalon FAQ
Tap a question to expand only the answer you want to read.
Is Epitalon FDA-approved?
No. Epitalon is not an FDA-approved medication in the United States, and there is no FDA-approved Epitalon product for longevity, anti-aging, insomnia, or other uses.
Is Epitalon the same as Epithalamin?
No. Epitalon is the specific synthetic tetrapeptide Ala-Glu-Asp-Gly, while Epithalamin is a complex pineal peptide preparation. Results from Epithalamin studies should not automatically be attributed to Epitalon.
Does Epitalon lengthen telomeres?
Epitalon has increased telomerase activity and telomere length in cultured human cells. Those findings are in vitro and do not establish that taking Epitalon lengthens telomeres in the human body or produces a health benefit.
Does Epitalon increase telomerase?
Cell-culture research has reported increased telomerase-related activity after Epitalon exposure. The clinical meaning of that effect in humans remains unknown.
Does Epitalon make people live longer?
There is no strong clinical evidence showing that Epitalon extends human lifespan. Longevity claims are largely based on cellular, animal, and older peptide-bioregulator research.
Does Epitalon improve sleep?
There is not enough evidence to establish Epitalon as an insomnia treatment. FDA's 2026 review concluded that available evidence does not support its effectiveness for insomnia.
Does Epitalon increase melatonin?
Some human and animal research has reported changes in melatonin or melatonin metabolites after Epitalon or related pineal-peptide exposure. These findings do not by themselves prove a clinically meaningful sleep benefit.
Is Epitalon safe?
Its human safety profile is not established. FDA has stated that adequate clinical safety data are lacking and has identified potential concerns involving immunogenicity, aggregation, peptide impurities, and product characterization.
Can Epitalon cause cancer?
There is no evidence proving that Epitalon causes cancer in humans. However, because Epitalon has activated telomerase in laboratory studies and telomerase is relevant to cancer biology, FDA has noted that available data are insufficient to fully assess carcinogenic potential.
Can pharmacies compound Epitalon?
FDA evaluated Epitalon-related bulk drug substances in 2026 and proposed against adding Epitalon free base and Epitalon acetate to the Section 503A Bulks List. Compounding law is specific and can change, so an online product should not be assumed to be an authorized compounded drug.
Why are Epitalon products difficult to compare?
The common name Epitalon has been used for different salts and related materials. Without validated analytical testing, labels alone do not establish identity, purity, concentration, or equivalence to research material.
How do you calculate Epitalon reconstitution concentration?
Divide the labeled amount in the vial by the liquid volume. For example, 10mg divided by 2mL equals 5mg per mL. This is concentration math rather than a recommendation for how Epitalon should be prepared or used.
Can the BACToBasics calculator handle Epitalon math?
Yes. Enter the labeled vial amount and liquid volume to calculate concentration, equivalent liquid volume, U-100 syringe units, and approximate amounts per vial. BACToBasics provides educational math rather than medical or dosing advice.
