What is gonadorelin?
Gonadorelin is synthetic GnRH, the same 10-amino-acid hormone released by the hypothalamus to control the reproductive axis. GnRH binds receptors on pituitary gonadotroph cells and triggers release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH).
Was gonadorelin FDA-approved for humans?
Yes historically. FDA records show human gonadorelin hydrochloride (Factrel) and gonadorelin acetate (Lutrepulse) products with approved NDAs, but both appear in FDA's discontinued drug-product listings. That historical approval should not be confused with the current veterinary Factrel product or with unapproved online research-market gonadorelin.
Human diagnostic use
Historically, gonadorelin was used diagnostically to evaluate hypothalamic-pituitary reproductive function by measuring the pituitary LH and FSH response to GnRH stimulation. Current U.S. human availability differs from that historical regulatory context because the former human products are discontinued.
Pulsatile GnRH and reproductive physiology
GnRH physiology is strongly pulse-dependent. Human studies using pulsatile GnRH have demonstrated restoration of pituitary-gonadal signaling, ovulation, pregnancy, testosterone production, and spermatogenesis in selected patients with hypothalamic or congenital hypogonadotropic disorders. Those studies involved medically supervised pump-based treatment, not generic online peptide protocols.
Why pulse pattern matters
The reproductive axis responds differently to pulsatile versus sustained GnRH receptor stimulation. Physiologic pulses promote LH and FSH secretion, while continuous stimulation can lead to receptor desensitization. This is one reason gonadorelin should not be reduced to a simple 'raises testosterone' claim.
Gonadorelin vs kisspeptin
Gonadorelin is GnRH itself and directly stimulates pituitary GnRH receptors. Kisspeptin acts one step upstream, primarily stimulating endogenous GnRH signaling through KISS1R. In a direct human comparison, GnRH produced a greater gonadotropin response than kisspeptin-10 or kisspeptin-54 at the tested intravenous doses.
Current veterinary gonadorelin is not a human product
DailyMed currently lists FDA-approved gonadorelin products such as Factrel for veterinary use in cattle. Veterinary approval, concentration, labeling, and manufacturing status do not make those products approved for human use.
Anti-doping status
The 2026 WADA Prohibited List explicitly names GnRH/gonadorelin and GnRH agonist analogues under S2.2.1 testosterone-stimulating peptides in males. Competitive athletes should verify current anti-doping rules before using any reproductive-axis hormone or peptide.
